<Desc>Science Overview for 2025 and the Global Problem We Seek to Solve1) It is believed by many that the global crisis of homeless companion animal overpopulation, the number one cause of suffering for dogs and cats, increased throughout 2025 as their primary source of food increased: human garbage. Millions of homeless dogs and cats continued to suffer as the cycle of reproduction of the homeless by the homeless repeated generation after generation. To end this suffering, our mission remained science-driven and unprecedented: to end the Number One cause of *continued on Sch O *continued from page 2* suffering and premature death for dogs and cats worldwide -overpopulation - by advancing the development of what will become the first one-dose, edible, permanent-lasting birth control formula capable of safely sterilizing stray dogs and cats without surgery.Recognizing that traditional spay and neuter surgery alone cannot solve this problem worldwide, we continued advancing the development of the Spay and Neuter Cookie as a cruelty-free, scalable, science-based, effective alternative. Building upon prior progress, we continued testing chemicals to produce lifelong sterilization from a single cookie, while our small dedicated team continued advancing this groundbreaking and complex scientific work, despite serious constraints ranging from threats and harassment to insufficient resources and funding. 2) In 2025, we were compelled to spend a substantial amount of effort improving the crucial analysis of 'the follicle count'. Before delving into these specifics, it is important to understand why the follicle count is so important. In short, the follicle count tells us whether our birth-control formula is actually working. Follicles are located in the ovaries, and once a follicle dies (usually with advancing age), it cannot be replaced, and without enough follicles, reproduction cannot take place. This natural condition in humans is referred to as menopause. As our goal is to develop a single-dose, permanent, lasting birth control treatment for dogs and cats, the most important question is: Did our formula permanently shut down the ovary's ability to produce future eggs? The follicle count helps answer that question. Before the follicle count: Researchers often had to rely on indirect clues: Did the animal become pregnant? Or did hormone levels change? Any number of questions. The problem is that these clues can be misleading. The follicle count gives us a more direct answer. By carefully examining treated ovarian tissues and analyzing their follicles, we can determine: How many follicles remain. What types of follicles remain? Whether the treatment damaged only some follicles or nearly all of them, and so on.Why this matters: We are not trying to create a birth control method that works for a few months. We are working to develop something far more ambitious: A safe, inexpensive, edible, one-dose treatment that permanently prevents reproduction - without the need for capture, injections or surgery. To prove that such a treatment truly works, we need evidence that the formula has eliminated, or sufficiently depleted, the viable ovarian reserve. The follicle count is one of the strongest ways to demonstrate that. A robust follicle-count method can help answer questions from: Scientific reviewers, Grant agencies, Regulatory authorities, Veterinary experts, and Potential partners. It provides objective evidence rather than relying on assumptionsIn short: The follicle count is important because it allows us to directly measure whether our one-dose formula has permanently disabled the ovary's capacity to produce fertile eggs that are necessary for becoming pregnant - the central requirement for ending the uncontrolled reproduction in stray dogs and cats.3) Analyzing the Follicle: Our Attempts to Automate Pathology Reporting with Artificial Intelligence. In 2025 we invested significant time in attempting to automate our pathology reporting using AI, hoping to speed up the expensive, labor-intensive work of evaluating follicles digital slide by digital slide. Unfortunately, this effort was not successful. AI-based classification and analysis is simply not good enough yet to meet our needs. Unfortunately it does not yet meet the level of accuracy that our work demands. In short, the variability and subtlety that we require, in follicle detection and staging, continued to exceed what current automated tools could/can reliably handle. While the experiments did not deliver the time savings we had hoped for, they clarified where automation can and cannot yet be trusted, and it reinforced why our rigorous, human-led pathology, and tools like our follicle quiz, remain essential. 4) To deal with this important problem, we developed the follicle quiz pipeline - the follicle quiz for short. Much effort had to be invested in developing the follicle quiz. The need for it grew directly out of a problem we kept running into: we observed real variability and differences in follicle detection and classification - across different veterinary pathologists, and there was no easy way to understand or quantify or localize where those disagreements were coming from or what they were based on. As the accuracy of each formula depends on how reliably follicles are counted and staged, this kind of variation is not something we could, nor can, afford to leave unresolved. Static annotations and side discussions were and are, not enough. We needed a way to directly visualize and compare how different pathologists were interpreting the same raw material: treated follicles. This is why we had to build a structured comparison tool ourselves.Although the result appears to be a single, rather simple yet highly structured product, in the form of an online platform, getting there required a deeply involved process, requiring several structured, interconnected steps. We first evaluated annotation software such as ImageScope and QuPath, and after careful testing and comparison we found QuPath to be more suitable and easier to build a workflow around, and therefore decided to proceed with it. We then turned to image extraction and processing: to enable analysis at the level of individual follicles, one by one in a format easy for both pathologists and non-pathologists to work with. To accomplish this we also needed to convert each annotated follicle into a standalone image, which required developing a pipeline to extract annotated regions (of digital histological slides of our treated ovaries) from whole-slide images and save them as individual online files. Finally, we built the follicle quiz platform itself, both frontend and backend interactively refining it for functionality and usability, integrating the image processing pipeline with the quiz system, and carrying out multiple rounds of testing to ensure correctness, stability, and reliability. In other words we built an online platform that allows multiple pathologists to all examine the same 100 to 1,000 treated follicles, at the same time, and analyze each follicle at the same time, in a user friendly much more time efficient manner than ever before, and ultimately, deliver the combined data to us in format that makes it much easier to compare and understand the analysis by each pathologist individually and as a group. 5) Digital Slide Quality and Proper Ovarian Histology: We discovered that not all of the digital slides were quite up to standard, and we decided to search for companies capable of preparing them at the highest level. Slide quality is not a minor detail, it determines whether our follicle counts are meaningful. To properly evaluate the treated ovary, the objective is to obtain a true cross-section that includes all key anatomical components, and in their correct spatial relationship. The follicles themselves are stratified by developmental stage: for example, the most immature primordial and primary follicles are typically clustered in the outermost region of the cortex, just deep to the capsule, while secondary and tertiary follicles are found deeper within the cortex as they mature. To continue to accurately assess the follicular population, for example for follicular counts or staging , the histological slide must represent this complete architecture. We spent considerable time getting this right, because without it, even a perfectly designed formula may not be reliably evaluated, which of course is unacceptable. 6) As to be expected, we encountered some related problems which we also had to resolve, for example in the case of Pilot Pup Allie. Her treated tissue on the slide was problematic because the section appeared to miss or underrepresent the follicle-bearing cortical region, possibly due to tissue orientation within the cassette or sectioning near the pole of the ovary. We deemed the slide as essentially non-diagnostic for our follicular analysis</Desc>

<ExplanationTxt>needs because the section did not include enough of the cortical tissue where the follicles are located. In the same group (of 4) as Allie, the slide from Pilot Pup Lila was perfect - though the 3rd and 4th slides both had issues with orientation of the tissues within the cassette, but with slightly different outcomes. Solving this required us to deliver more explicit instructions to the histology processing companies, which they then needed to carry out. 7) Another problem we had to find a solution to is illustrated by the case of Pilot pup Abbey, where her tissue was sectioned on a tangential or oblique plane, rather than a true cross-section. The blade appeared to have passed through the ovary at a more shallow, non-perpendicular angle. The section appeared to include a larger proportion of the deeper cortex. This creates a distorted view that artificially enriches the population of secondary follicles while simultaneously missing the outer cortex where the primordial/primary follicles reside. The slide was not representative of the true follicular distribution; it created a misleading artifact of follicular dynamics by selectively over- and under-representing different zones. In this instance, the anatomical integrity of the cortex-medulla relationship was not fully captured, preventing a valid histopathological analysis of the follicular population. Again, solving this required us to deliver more explicit instructions to the histology processing companies, which they then needed to carry out. 8) In 2025, we of course continued advancing our evaluation of reproductive blockers and ovotoxicants - while continuing to develop formulations to meet the six criteria we deem to be essential to produce a practical, edible, cruelty-free and scalable non-surgical sterilization solution to the global overpopulation problem, starting with dogs and cats before advancing to other species in need. We were able to increase our understanding of ingredient interactions and outcomes, including evaluating promising new chemical candidates, with compound discovery work through good old fashioned scientific review and database research. 9) We also continued advancing active ingredient optimization strategies (as in developing new formulations) to improve single-dose effectiveness - while staying within self imposed boundaries, for safety and to remain cruelty-free. This included integrating multiple active and inactive ingredients into single-dose formulations to improve three crucial elements: Ingredient compatibility. Product stability. Product integrity. Including: preserving effectiveness under real-world conditions - which is easier said than done. 10) International vs. U.S. Trials: We continued advancing our research in cost-effective international settings, with the goal of saving enough funds to transfer into the U.S. to pursue the FDA and USDA processes necessary for deployment of the future Spay and Neuter Cookie, in the U.S. 11) Outreach and Advocacy: We continued advancing our outreach and advocacy efforts to raise awareness of animal suffering beyond overpopulation, and encourage public action on behalf of suffering animals worldwide regardless of the cause of their suffering. We continued educating and mobilizing our supporters and subscribers (approaching 50,000) through digital communications and advocacy initiatives, to fight cruelty to all animals.</ExplanationTxt>
